MS solid medium is widely used in plant tissue culture for inducing callus formation or for culturing embryos, stems, shoot tips, and anthers. Its liquid form is ideal for cell suspension cultures and has shown significant success. The inorganic nutrient content in MS medium is carefully balanced to meet the physiological and nutritional needs of plant cells. As a result, it's generally unnecessary to add organic components such as amino acids, casein hydrolysate, yeast extract, or coconut water.

A notable feature of MS medium is its higher levels of nitrate, potassium, and ammonium compared to other media like B5. In contrast, B5 medium contains lower ammonium concentrations because ammonium can sometimes inhibit plant growth. Experimental results show that some plant callus and suspension cultures grow better on MS, while others thrive more on B5. This makes the choice of medium highly dependent on the specific plant species and experimental goals.

N6 medium is particularly suitable for anther and pollen culture in cereal plants and is commonly used both domestically and internationally. Other frequently used media include White (1963) and Nitsch (1951) media, which share similar basic compositions. The White medium is preferred for woody plants due to its lower inorganic salt content.
